This hand book details the process of Cataractogenesis using 'Lens Organ Culture Technique' and the changes in the lens enzymes. Three models were selected viz sugar induced cataract, hydrogen Peroxide induced cataract and steroid induced cataract. A total of 480 Goat lenses, obtained from slaughter house were analyzed. These lenses were subjected to 'Lens Organ Culture Technique'. The cultured lenses were categorized as control group and the experimental groups (Sugar induced cataract H202 induced cataract and steroid induced cataract). It was found that total soluble proteins were significantly decreased in all the three types of experimental cataractous lenses when compared with control lenses. The activity of catalase did not show any significant decrease in sugar cataracts and steroid cataract while a significant decrease was observed in H202 cataract as compared to control lenses. Specific activities of Glutathione peroxidase and Glutathione reductase were found to be significantly decreased in all the three types of cataractous lenses as compared to control lenses. The activity of other enzymes was also measured (lactate dehydrogenase, Glucose-6-phosphate dehydrogenase).
