Master's Thesis from the year 1999 in the subject Biology - Genetics / Gene Technology, grade: 11 on 13-scale, Technical University of Denmark (Microbiology), language: Danish, abstract: Dette projekt handler om yutK genet i Bacillus subtilis. Formålet med projektet var at undersøge den konstruerede WOT 22 (yutK :: pMUTIN4) stamme, som var blevet fremstillet i forbindelse med mit 15 points forprojekt. Denne stamme skulle analyseres og sammenlignes med Bacillus subtilis vildtype stammen. Der var blevet konstrueret 10 kontrol stammer, alle indeholdende en transkriptionel fusion mellem et fragment fra yutK indeholdende dettes operator -/ promoter område og lacZ, indsat i amyE. lacZ genet koder for beta-galactosidase. Alle disse stammer samt WOT 22 blev analyseret ved fænotypisk ekspressions undersøgelser, og der blev målt beta-galactosidase-aktivitet af yutK stammen (WOT 22) og kontrol stammen WOT 268(3) i 7 punkter på vækstkurven.
Resultaterne viste, at yutK stammen (WOT 22) havde h.h.v. 2,45, 0,79 og 0,90 fold lavere beta-galactosidase aktivitet fra eksponentiel til stationær fase, når stammen voksede på flydende h.h.v. LB medie og minimal medie med h.h.v. glucose og succinate samt glutamate tilsat. Imens kontrol stammen WOT 268(3) havde h.h.v. 8,12, 1,47 og 2,06 fold lavere beta-galactosidase aktivitet fra eksponentiel til stationær fase, når stammen voksede på flydende h.h.v. LB medie og minimal medie med h.h.v. glucose og succinate samt glutamate tilsat. Resultaterne i dette studie viste, at vildtypen blev hæmmet, når yutK genet blev slået ud, og at yutK genet var omvendt catabolite repression.
Sekventering af B. subtilis PCR-produkter, som var opformeret fra kontrol stammerne WOT 268(3) og -(8) viste, at PCR-produktet fra WOT 268(3) stammen ikke indeholdte nogen afvigelser fra den ønskede sekvens. Widad Akrawi is a geneticist and global health expert, and known for being a Danish human rights defender, peace advocate, and author. She was born 1969 in Akre, Kurdistan region. She graduated as a civil engineer in 1990, and proceeded on to the Technical University of Denmark where she earned a Masters degree in genetics and microbiology. After graduation, she served as a clinical geneticist and researcher in human genetics and inherited diseases at the Danish Royal Hospital, the largest hospital in Denmark and a leading university hospital in Northern Europe. She has also earned a PhD degree in global health and epidemiology.In 2003, she became a published writer. After researching the concept of victim psychology and the effects of various types of trauma on individuals' psychological health, she published her first novel, Tara's Book, written in Danish, in Copenhagen. The book was translated to Norwegian in 2005, published in Oslo, and distributed by the Norwegian Cultural Council to almost all Norwegian libraries. Her eBook, entitled Under Hbets Skyggee (Under the Shadow of Hope), also in Danish, was published in 2013 at Amazon.com and Lulu.com. She currently leads Defend International, a Norway-based NGO that is devoted to medical research, and human rights. Over the years, she has addressed and actively participated in several regional and international meetings and conferences, such as the United Nations Third and Fourth Biennial Meeting of States, the UN Commission on The Status of Women, the General Assembly Interactive Hearings with Non-Governmental organizations, Civil society organizations and the Private sector, and the UN First Committee on Disarmament and International Security. She has held numerous leadership positions in national and international organizations, including executive board member of Amnesty International Denmark, ambassador for Amnesty International's "Stop Torture" campaign, and member of the Panel of Vestby Experts on approaches to socio-cultural constructivism. At present, she is a board member of Women of Europe Award, an active member of International Action Network on Small Arms (IANSA), a board member of IANSA Women's Network Working Group, and a senior lobbyist at the UN, lobbing for the adoption of an international Arms Trade Treaty during the negotiations that take place at the UN General Assembly's First Committee on Disarmament and International Security.
